I don't think DeSantis will overplay his hand by trying to take the Republican nomination from Trump when doing so could only end in disaster for him. All he could feasibly end up doing is pissing off Trump, costing himself a potential VP spot and his reputation with MAGA voters. He's only playing coy about it because he doesn't want to look subservient to Trump and he will still probably run if Trump chooses not to.
