We do have a quite a few here in Germany and they do actually work. Tough we do also femce of most of our highways, so that might also help.

Animals aren't dumb. They will notice trends and the fact that other animals not using this system are being killed.

If you wall it off, no animals can across the highway and than you start getting isolated pockets of fauna. To prevent that you thrn build green bridges. Also you can't wall off the entire highway, so you'll funnel a lot of animals into a few chokepoints if you dont have any save for them cross.
