We don't use since+ago in the same sentence. So the preposition here is not necessary. "my neighbors moved in over (more than) three years ago" is another possible answer, but changes the meaning slightly.

We don't really say that, for the same reason as before. Left is a completed action, while since is a continuous action. "My sister has been gone since last Saturday" would be correct. Because that uses a continuous verb. So I think the correct answer here again is "not necessary".
