DC literally has only 5 notable female villains. Even then, characters like Harley/Ivy/Selina are mostly anti-heroes now.
So, how should they improve their villain cast? Make one of the unused heroes into a villain. Power Girl i think would be the perfect pick, since the idea of a being as powerful as a kryptonian falling for and serving the clown prince of crime would be amazing.
She should be reintroduced as a broken hero. Barely able to tap to her former potential but this time she's not holding back. It also, would be a perfect way to reintroduce joker since he's been so overused for the past decade. Since, now he has to share the spotlight with his new bitch.
There was a porn comic by the last pharoah I think where wonder woman gets jokerized.

It's better than letting the hero be reduced to cameo appearances. They tried replacing her in the new 52 and now she only appears in JSA cameos.
Making her a villain will reignite interest for the character and make people want to read if she'll ever come to her senses again.

Power Girl already had a comic run with Harley. There's approximately zero chance she wouldn't have a bone to pick with Joker if he tried anything.
If you're insistent on a Batman-themed evil villainess arc, your better bet would be Poison Ivy. She's controlled Superman with kryptonite lipstick kisses before, so there's at least a precedent. Maybe have a couple issues of her doing villainous acts, confusing the heroes. Harley, of all people, figures it out and has to save the day. You can even do some shit where Ivy was trying to solve some issue with pollution or industrialization and only gets a slap on the wrist to restore status quo.
Or make an actual rouges gallery for PG. She has a bunch of one-offs that exist for very brief periods of time.
That was a clone of Super Girl, and the animators of JLA continued to be groovy dudes and made her a reference to Power Girl, visually.

Circe is the only one who gets used relatively frequently and relatively well. Everybody else doesn't show up or when they do they either job or the writer tries to them into antiheroes or antivillains.
