-There's like 20 different kinds of K including one that turns someone romantic or gay.
-Many of the old pulp era stories, including Sherlock Holmes, were known for taking some kind of poison every day to build up an immunity to it.
-Different writers, different focus, different "Hey, wouldn't it be neat if X happened without explanation?"
-DC hasn't been using writers who can write Superman properly for decades. There's a point where it's on purpose and they've clearly passed it

It's a macguffin literally made to make it so the Superman Voice Actor from the Radio Show could go on vacation.
It's inconsistent like all macguffins because it's meant to facilitate a story in some form and is only beholden to the writers.

Nowadays, nothing aside from brand recognition. It's an artificial weakness created 80 years ago, back when the cape genre was much less developed and more primitive. It's been criticized for hell and back for not really adding anything interesting to Superman's character. But it's become so closely associated with the character that you can't just do away with it. And if DC dared retcon it, like made some big story where Superman stops being vulnerable to kryptonite, people would bitch that Superman is "even more OP now". And they would be partially right because the right way to go about it was to destroy kryptonite AND introduce or emphasize some sort of a more human, psychological weakness to Superman. But a) it would be pretty jarring to suddenly introduce a new trait like that, b) you can't make a big news headline out of "Superman is going to be anxious about X now", c) Superman already has traits like this, like his unwillingness to cross lines and change the world on a greater scope, but they often aren't treated like weakness and people don't care about them.

If they didn't have krypronite, what would the artists have used? Kryptonite could turn everything green, in an eerie light sort of way. And as much green as they could had to be used, since green was the cheapest ink. Without it, would Supes have had a green costume?
