Dave is one of those "practicing and music theory will kill your individuality" guys. I remember seeing a video of him talking about the guitar lessons he had to take after his arm injury and getting mad at the teacher for correcting his "bad habits" because he thought it was gonna ruin his style.

you need to listen to more metal btw

style and guitar tone, but mostly style. in some bands with two guitar players it can be harder or easier to distinguish the two, but with Megadeth it's usually easy since Dave has a very simple, "Jimmy Page and Ace Frehley on steroids" while his guitarists are in a more neoclassical or jazzy style.
also helps to look up live performances to see who plays what
